Testing a power supply manually with a multimeter is one of two ways to test a power suin a computer. A properly executed PSU test using a multimeter should confirm that the power supply is in good working order or if should be replaced.
In 20 pins conn., uses pin 14 (power on line, green) loops to common ground (black) together.
In 24 pins conn,, uses pin 16 (power on line, green) loops to common ground (black) together.
